Thanos (Josh Brolin) wie auch Quills Vater Ego (Kurt Russel) sind zwar Geschichte, aber dennoch sind die hinterlassenen Schäden sowohl im Universum als auch bei den Guardians nach wie vor spürbar. Um den gigantischen Schaden, den Thanos angerichtet hat, zu reparieren, wird ein neuer Zufluchtsort geschaffen. Nachdem sie dem Collector Knowhere abgekauft haben, soll dies fortan als eine Stätte des Friedens gelten.
Von der rachsüchtigen Hohepriesterin Ayesha (Elizabeth Debicki), ihrer Kreation Adam Warlock (Will Poulter), aber auch dem mächtigen High Evolutionary (Chukwudi Iwuji), ahnen die Guardians zu dem Zeitpunkt allerdings noch nichts. Als der Frieden eines Tages gestört wird, müssen sie erst einmal herausfinden, wer Freund und wer Feind ist. Erneut haben Star-Lord (Chris Pratt), Gamora (Zoe Saldana), Groot (Stimme im Original: Vin Diesel) und Co. alle Hände voll zu tun, um den Frieden aufrechtzuerhalten.
